摘要:topicalexposuretonanoscalematerialsislikelyfromavarietyofsourcesincludingsunscreensandcosmetics.becausetheinvivodispositionofnanoscalematerialsisnotwellunderstood,wehaveevaluatedthedistributionofquantumdots(qds)followingintradermalinjectionintofemaleskh-1hairlessmiceasamodelsystemfordeterminingtissuelocalizationfollowingintradermalinfiltration.theqd(cdsecore,cdscapped,poly[ethyleneglycol]coated,37nmdiameter,621nmfluorescenceemission)wereinjectedintradermally(id)ontherightdorsalflank.withinminutesfollowingintradermalinjection,thehighlyuvfluorescentqdcouldbeobservedmovingfromtheinjectionsitesapparentlythroughthelymphaticductsystemtoregionallymphnodes.residualfluorescentqdremainedatthesiteofinjectionuntilnecropsyat24h.quantificationofcadmiumandseleniumlevelsafter0,4,8,12,or24hinmultipletissues,usinginductivelycoupledplasmamassspectrometry(icp-ms),showedatime-dependentlossofcadmiumfromtheinjectionsite,andaccumulationintheliver,regionaldraininglymphnodes,kidney,spleen,andhepaticlymphnode.fluorescencemicroscopycorroboratedtheicp-msresultsregardingthetissuedistributionofqd.theresultsindicatedthat(1)idinjectednanoscaleqdremainedasadepositinskinandpenetratedthesurroundingviablesubcutis,(2)qdweredistributedtodraininglymphnodesthroughthesclymphaticsandtotheliverandotherorgans,and(3)sentinelorgansareeffectivelocationsformonitoringtransdermalpenetrationofnanoscalematerialsintoanimals.
